600个明星的电话号码曝光后，偷窥迷糊精易淑（周笔畅）的电话号码因与其中之一大明星天佑（张柏芝）的仅相差一位，偷窥成为“第601个电话”，平静简单生活自此变成啼笑皆非的闹剧。就在她不胜其烦要换号码时，接到身患重病的大男孩阿文（胡歌）的电话，听过阿文的故事，易淑决定抛弃原有的生活去寻找天佑，帮助阿文达成心愿——希望听到心爱的偶像天佑演唱自己写的一首歌。而站在绚丽舞台上的天佑，在收获鲜花和掌声的同时，有着常人无法理解的压力，她在等待一个机会，来像易淑一样下定决心改变自己的旧生活。
